Abstract

Metastatic triple-negative breast cancer (mTNBC) is an aggressive subtype of breast cancer associated with poor prognosis and limited therapeutic options, prompting the development of targeted therapies such as Sacituzumab Govitecan, a Trop-2-directed antibody-drug conjugate. This study aimed to evaluate the efficacy of Sacituzumab Govitecan in patients with mTNBC through a systematic review and meta-analysis of randomized controlled trials. A comprehensive literature search was conducted in PubMed, Embase, and the Cochrane Library up to December 2024 following the PRISMA guidelines. Randomized controlled trials comparing Sacituzumab Govitecan with standard treatment or placebo and reporting overall survival (OS) and progression-free survival (PFS) were included. Data were analyzed using a fixed-effect model, and treatment effects were expressed as hazard ratios (HRs) with 95% confidence intervals (CIs). Four randomized controlled trials involving 1,383 participants, including 700 patients receiving Sacituzumab Govitecan and 683 receiving control treatment, were included in the analysis. Sacituzumab Govitecan significantly improved overall survival (HR = 0.51; 95% CI: 0.45–0.58; p < 0.00001) and progression-free survival (HR = 0.41; 95% CI: 0.36–0.47; p < 0.00001) compared with control, with no significant heterogeneity among studies (I² = 0%). These findings demonstrate that Sacituzumab Govitecan provides substantial survival benefits and represents an effective treatment option for patients with metastatic triple-negative breast cancer.
